Medium "Cancirost" with glutamine

Ready-to-use medium "Carcinost" is intended for obtaining primary cell cultures of tumor neoplasms in vitro for cytological, cytogenetic and molecular biological studies. The medium provides selective proliferation and clonal growth of poorly differentiated cells of solid tumors of various origins. Indicators of proliferation and differentiation of colony cells can be used in the diagnosis of the analyzed neoplasms malignancy degree.
The sequence of procedure for obtaining primary cultures of tumor tissues.
METHOD OF APPLICATION: Ready for use medium. It contains a mixture of broad-spectrum antibiotics of antibacterial action. For sterile work open the vial should be under aseptic conditions.
Before use, mix the contents of the vial with the "Carcinost" medium (especially in the case of using a defrosted product).
5 ml of the medium is added into a culture vial of 25 cm2 and placed in a CO2 incubator (5-6 vol. % CO2, 90-100% humidity, 30 - 37 °C).
Seed suspension of tumor tissue is prepared by mechanical or enzymatic disintegration of the sample (for operating samples 0.5-1.0 cm3)*.
0.5- 1.0 ml of the resulting suspension is added into a preincubated vial with "Carcinost" medium and incubated for 48-72 hours to attach adhesive elements.
Completely remove the medium with the seed material and add 5 ml of fresh medium.
Cell culture is carried out in a CO2 incubator for 1.5 - 4 weeks (depending on the test sample material clonogenicity level) **.
* In order to avoid damage to cellular material and protein components of proliferative medium, it is preferable to use mechanical method of tissue disintegration with crushing into fragments up to 1-3 mm3 and subsequent shaking on a vortex for 1-1.5 minutes in 2-3 ml of isotonic solution.
** The medium volume is changed during cultivation every 4-7 days.
HOW SUPPLIED: in hermetically sealed plastic bottles (PET containers) of (60 ± 1.5) ml.
Shelf life: 1 year.
Storage and transportation conditions: Store in a place protected from light at temperature of -10 ...-20 ° C.
